ALK and non-small cell lung carcinoma: In two randomized phase III trials, crizotinib was associated with significantly higher PFS (10.9 months vs. 7.0 months in the first-line PROFILE 1014 trial and 7.7 vs. 3.0 months in the PROFILE 1007 trial involving pre-treated patients) and objective response rate (ORR) compared to first- and second-line cytotoxic chemotherapy, resulting in its approval as frontline therapy in patients with advanced, ALK-positive NSCLC[9,13].
