Recently, a PTK7 germline variant bearing a missense mutation substituting valine 354 into methionine (V354M) within the fourth Ig-like loop was identified in familial colorectal cancer (CRC) cases and found associated to the oncogenic function of PTK7 by increasing cell proliferation, migration, and invasiveness (Fig. 3) [21]. This evidence concerns the gene PTK7 and familial colorectal cancer.
