In contrast, the O-GlcNAcylation of STAT3 has been shown to be detrimental in conditions such as inflammatory bowel disease and inflammation induced colon tumorigenesis; its O-GlcNAcylation at T717 inhibits STAT3 Y705 phosphorylation resulting in the loss of IL-10 production in macrophages, leading to exacerbated colon inflammation (32).
