In particular, the rTg4510 mouse model is now known to exhibit off-target mutations around the site of MAPTP301L transgene insertion that may contribute to pathological phenotypes.80,81 However, studies that have used doxycycline to suppress transgene expression at different developmental age points in rTg4510 mice have shown a range of tauopathy-driven cellular and synaptic phenotypes.42,82 Overall, the rTg4510 mouse is one of only a few well-characterized models that display robust tau pathogenesis and neurodegeneration.
