NSD2 and cancer: Recently, NSD2 degraders, a new type of NSD2 modulatorthat couldabolish the catalytic and noncatalytic functions of NSD2, have beenreported (Figure 1).In 2022, Jin and co-workers reported a first-in-class NSD2 degraderMS159 (6) by employing proteolysis targeting chimera(PROTAC) approach.19 While compound 6 dose-dependently degraded NSD2 and was more effective insuppressing the growth of cancer cells than the parent NSD2 binder,its degradative activity on NSD2 was relatively low (DC50 = 5.2 μM in 293FT cells after 48 h treatment).
