B2R is an essential G protein-coupled receptor(GPCR) involvedin regulating homeostasis of the cardiovascular system as a vasodepressor.4 Binding of bradykinin to B2R promotes vasodilation,increased endothelial permeability, and capillary bed leakage, whichin severe cases of bradykinin overproduction displays the symptomsof an acute angioedema attack.5 Accordingly,significant interest has developed around PKa as a drug target forthe treatment of bradykinin-mediated angioedema such as hereditaryangioedema (HAE).
