Indeed, whereas overexpression of wild-type FOXO1 significantly enhanced the proportion of CD45RA+ and CD62L+CD27+ CAR T cells (Fig. 3a)—in contrast to FOXO1-ADA expressing CAR T cells—wild-type FOXO1-expressing CAR T cells were able to produce similar levels of IFNγ and TNF to control CAR T cells upon co-culture with tumour cells (Extended Data Fig. 7e).
