In 2019, researchers led by Roland Imle (17) confirmed the localization of ADPGK within the endoplasmic reticulum, and illustrate that ADPGK is part of a glucose sensing system in the ER modulating metabolism via regulation of N- and O-glycosylation, ADPGK deficiency resulted in reduced glucose uptake, diminished activities of hexokinase, phosphofructokinase, and respiratory chain complexes, activation-induced depletion of thymidine metabolism intermediates, and enhanced activation of autophagy, ultimately leading to a severe energy metabolism disorder.
