IDH1 and cancer: IDH1 inhibition has been proved to enhance the effects of classic cancer therapies in several types of cancer by decreasing αKG, ATP, lipid, and dNTP synthesis and increasing ROS, lipid peroxidation, and protein oxidation, all of which are closely related to the IDH1 enzymatic activities.[22] These findings suggest that activated IDH1 may mitigate the damage caused by stress that increases cell death and that inhibition of IDH1 activity may have therapeutic potential in some diseases, including cancer.
