PKMYT1 and cancer: PKMYT1 knockout or RP-6306 treatment inhibits cell cycle progression (Figs. 3H, 4D, and EV2F) and induces pan-γH2AX in PDAC cancer cell line (Figs. 3I,  4C, and EV2C), which indicate that G2/M checkpoint dysfunction caused by PKMYT1 inhibition leads to unchecked premature mitotic entry, then results in the accumulation of genetic lesions from unrepaired DNA damage, ultimately leading to apoptosis or mitotic catastrophe (Figs. 3J,  4E and EV2G) (Asquith et al, 2020; Gallo et al, 2022).
