As expected [6,7,17,21,39], MX2 with a deletion of the N-terminal 25 amino acids (ΔN25), mutation of the N-terminal triple-arginine motif (RRR11-13AAA), or phosphomimetic S14D/S17D/S18D (SSS14,17,18DDD) did not inhibit HIV-1WT infection, while non-phosphorylatable S14A/S17A/S18A (SSS14,17,18AAA) had antiviral activity similar to WT MX2, that was abrogated in the presence of CsA in HT1080, but not HeLa cells (Figs 3, S6, and S7).
