Transgenic expression of CD33 in lung cancer cells that do not endogenously express CD33 (Supplemental Figure 3B) resulted in rapamycin-dependent DARIC-VHH1 T cell proliferation in vitro, whereas targeted deletion of CD33 from CD33+ AML cell lines eliminated IFN-γ responses in vitro (Supplemental Figure 3, C and D) and in vivo (Supplemental Figure 3G).
