MYH14 and myopathy: Because the core contact triad that interacts with actin residue E334 is present in all class 2 myosin isoforms, it is reasonable to assume that mutations of residue E334 in α-skeletal actin affect the interaction with skeletal muscle myosin-2 to a similar extent, resulting in less efficient force generation and thereby contributing to the development of myopathy in patients.
