In a lung cancer cell line, simultaneous administration of the irreversible tyrosine kinase inhibitor (TKI) neratinib and the antibody-linked drug T-DM1 has been demonstrated to increase HER2 internalization and enhance anti-tumor activity, while the reversible HER2 inhibitor lapatinib resulted in reduced endocytosis [11], suggesting that the mechanism for reversible and irreversible HER2 inhibitors may differ.
