The I148M variant disrupts the phospholipase activity of patatin-like phospholipase domain-containing 3 (PNPLA3), a triglyceride lipase, thereby interfering with lipid catabolism and promoting hepatic fat accumulation to increase the risk of liver-related mortality in patients with NAFLD [17, 18]. Here, PNPLA3 is linked to metabolic dysfunction-associated steatotic liver disease.
