Of the other two patients presenting with non-amnestic Alzheimer’s disease, one (case 2) had elevation of cerebrospinal fluid (CSF) total tau and phosphorylated tau; brain-restricted postmortem examination showed non-specific Aβ (diffuse deposits with patchy distribution restricted to the neocortex; single cortical blood vessel with concentric mural Aβ deposition) and tau deposition not meeting pathological criteria for Alzheimer’s disease (Fig. 2, Supplementary Information and Extended Data Fig. 1). The gene discussed is MAPT; the disease is early-onset autosomal dominant Alzheimer disease.
