Tumor-derived DNA activates cGAS to produce 2′,3′-cGAMP, the endogenous ligand of STING, resulting in the downstream signaling cascade via the recruitment of threonine-protein kinase (TBK1), phosphorylation of the interferon regulatory transcription factor IRF3, and production of type I interferon (IFN), and other proinflammatory cytokines.
