Inhibitors of PORCN are under active development for the treatmentof neurological disorders and cancers, with inhibitors WNT974 andETC-159 in early phase clinical trials.3−5 Inhibition of GOAT bypeptide-coenzyme A product mimetic GO-CoA-Tat, along with triterpenoid-basedinhibitors, indicate a potential route to target this MBOAT for therapeuticbenefit in type II diabetes and obesity.6,7 However, inhibitordevelopment programs targeting HHAT have shown more limited progressto-date. This evidence concerns the gene PORCN and obesity due to melanocortin 4 receptor deficiency.
