While the distribution of amyloid deposition in the brain early in the course of disease does not appear to be particularly informative about the likelihood of progression from normal cognition to MCI (Scheinin et al., 2009), tau tends to deposit in a systematic fashion early in the course of AD and then spreads in a relatively predictable manner to other brain regions (Pascoal et al., 2018). The gene discussed is MAPT; the disease is Alzheimer disease.
