In addition, we overexpressed the METTL3 in AsPC-1 and BxPC-3 cells by transfection with pEnter-METTL3 and demonstrated that celastrol-induced downregulation of Claspin and Bcl-2 was partially rescued by METTL3 overexpression (Fig. 5D-F), suggesting that METTL3 participated in celastrol-mediated regulation of Claspin and Bcl-2 in pancreatic cancer cells.
