Importantly, melanoma cells harbouring the BRAFV600E mutation are characterized by constitutive activation of the RAS-ERK signalling pathway, which promotes recruitment of ERK2/GABP or ERK2/ETS1 complexes on the mutant TERT promoter, phosphorylation of SP1, shift in HDAC1 occupancy, acetylation of histones H3K9, and reactivation of telomerase (Shi et al., 2015).
