Two novel drugs that represent a potential novel treatment to employ when myeloma is refractory to current therapeutic options are: selinexor, a selective inhibitor of exportin 1 (XPO1) [phase 2b STORM trial [65], and Iberdomide, a new IMiD whose mechanism of action consists in the degradation of proteins responsible for resistance (Ikaros and Aiolos) [phase I/II CC-220-MM-001 trial [66]]. The gene discussed is XPO1; the disease is plasma cell myeloma.
