When compared with vehicle-treated cocultures, the DAPT treatment significantly reduced the upregulation of WNT5B mRNA levels in WM852 and WM165 cells cocultured with LECs (Figure 5B and Supplemental Figure 6B), further supporting a key role for Notch3 in regulating WNT5B expression in the metastatic WM852 and WM165 melanoma cells.
