The observation that BRISC functions as a K63-specific DUB but has a positive role in the regulation of TLR4-NF-κB signaling pathway in KCs indicates that BRISC is unlikely to directly target the classic K63-linked polyubiquitinated proteins in NF-κB pathway, such as TNF receptor-associated factor 6 (TRAF6) and NF-κB essential modulator (NEMO). The gene discussed is TRAF6; the disease is dry eye syndrome.
