Collectively, this study highlights the critical role of METTL3 deficiency in the pathogenesis of allergic asthma airway inflammation, as featured by promoting M2 macrophage activation and enhancing Th2 response, and uncovers a signaling axis involving METTL3/YTHDF3-m6A/PTX3/STX17 in macrophage activation and autophagy maturation (Fig. 8f).
