The study aimed to: 1) identify proteins associated with risk of IHD independent of established cardiovascular disease (CVD) risk factors; 2) use cis-protein quantitative loci (pQTLs) identified in GWAS of CKB and UK Biobank (UKB) for these proteins to assess their causal relevance for IHD in 2-sample Mendelian randomization (MR) analyses in a global IHD GWAS consortium; and 3) explore mechanisms linking such proteins with IHD using enrichment analyses, tissue expression, and single-gene knockout (KO) models. This evidence concerns the gene CKB and myocardial ischemia.
