Androgen deprivation therapy (ADT), performed by castration (chemical or surgical) or by the administration of anti-androgen medications, is the mainstay of the treatment of advanced PC since the suppression of testosterone or blocking its connection to the AR (in the case of anti-androgens) results in fewer activated ARs and, consequently, a reduction in the transcription of the TMPRSS2 gene. Here, TMPRSS2 is linked to pachyonychia congenita.
