In another first-line therapy study (CheckMate-649), a significant advantage of combined immune chemotherapy vs. chemotherapy alone (FOLFOX or CapeOx) in the primary endpoint overall survival was demonstrated in HER2-negative adenocarcinoma of the stomach, the gastroesophageal junction or the esophagus: in PD-L1-positive tumors (CPS ≥ 5), the addition of nivolumab led to a median prolongation of survival to 14.4 vs. 11.1 months (HR 0.71; p < 0.0001) (Janjigian et al. 2021). This evidence concerns the gene ERBB2 and adenocarcinoma.
