When the five clinical risk factors—age, serum PSA, family history of PC, previous negative biopsy, and abnormal DRE—were added to the models, the combined Models I, II, III, and GS showed significantly improved AUCs (0.95, 0.92, and 0.92, and 0.87, respectively; all p < 0.0001), which were higher than those achieved by adding PSA alone to the models (0.94, 0.90, 0.90, and 0.85, respectively; all p < 0.05). The gene discussed is KLK3; the disease is pachyonychia congenita.
