Inhibition of the kinase activity of NPM::ALK or exposure to DNA methyltransferase inhibitors rescues the expression of CD3ε, ZAP70, LAT, and SLP76, suggesting that NPM::ALK-mediated transcriptional repression occurs via DNA methylation to downregulate components of the TCR signaling cascade in ALK+ ALCL (28). The gene discussed is ALK; the disease is anaplastic large cell lymphoma.
