PBMCs from each of the four donors (two with pancreatic cancer and two with colorectal cancer) were split into six separate peptide stimulation cultures each: four with individual mutant KRAS peptides (four of six mutant KRAS peptides were used due to limited sample availability, of which three corresponded to the most reactive peptides by ELISPOT [G12C, G12V, G12A] and one corresponded to the most reported TCR target [G12D]), one with a wild-type (WT) KRAS peptide, one with an Epstein-Barr virus (EBV) peptide, and one negative control culture without any peptide stimulation (Figure 2B).
