To investigate whether the increased number of O-linked glycosylation sites in CD43 affected the uptake of M. tuberculosis by macrophages, we transfected WT-SPN (wild-type, pCMV-WT-SPN) or HN-SPN (Hainan-type, pCMV-HN-SPN) into the RAW264.7 (ECACC 91062702) macrophage cell line 24 h before the infection by the H37Ra strain of M. tuberculosis at a range of multiplicity of infection (MOI) for 4 h (Fig. 3C).
