Small-molecule tankyrase inhibitors block Wnt signaling by stabilizing axin and axin2 (Huang et al., 2009), and next-generation tankyrase inhibitors are currently developed for targeted colorectal cancer therapy (Kim et al., 2022; Leenders et al., 2021), demonstrating the functional and clinical importance of tankyrase-mediated degradation of axin proteins.
