Nicotinamide phosphoribosyl transferase (NAMPT) is the apical and rate-limiting enzyme of the salvage pathway (Fig. 1A; Supplementary Fig. S1) and has been pursued as a cancer therapeutic target, as it is often overexpressed in cancer, and certain tumors exhibit absolute dependency on the salvage pathway due to lack of functional de novo or PH pathways (4, 5).
