To clarify whether the rescuing effects of CISD KD and KO are due to reduced IP3R activity, we observed PD-related phenotypes in PINK1 or Parkin null flies co-expressing CISD RNAi and exogenous Drosophila IP3R. While CISD KD and KO in PINK1 and Parkin null flies completely rescued PD-related phenotypes (Fig. 5a–f and Supplementary Figs. 9a–h, 10a–f, and 11a, b), simultaneous expression of IP3R resulted in recurrence of PD-related phenotypes, indicating that the rescuing effects of CISD KD and KO were strongly diminished (Fig. 5a–f and Supplementary Figs. 9a–h and 11a, b).
