Recently, the BEACON CRC trial, which enrolled patients with previously treated unresectable BRAF V600E mutant mCRC, demonstrated that the triplet regimen of encorafenib (a BRAF inhibitor), binimetinib (an MEK inhibitor), and cetuximab (anti-EGFR antibody) as well as the doublet regimen of encorafenib and cetuximab significantly improved both progression-free survival (PFS) and OS compared with standard chemotherapy (FOLFIRI/irinotecan plus cetuximab) [8, 9]. Here, BRAF is linked to colorectal carcinoma.
