Successful expression of the ion channel Piezo1 in HEK293T cells and subsequent Ca2+ influx triggered the downstream pathway for gene expression. The Piezo1 gene was transferred into Jurkat T cell lines and PBMCs to create chimeric antigen receptors, transactivated to open channels and stimulate ultrasound response. ReCoM is effective in controlling CAR expression in T cells to guide the recognition and eradication of tumor cells for controllable cancer immunotherapy.
