In all PF-3758309-treated parent myxofibrosarcoma cell lines relative to vehicle controls, PF-3758309 at 0.1 μM significantly abolished the levels of CFS2 mRNA (Figure-5D) and secreted CFS2 protein (Figure-5E), the PAK1 occupancy at the STAT5B binding site of the CSF2 promoter in the ChIP assay (Figure-5F), and the angiogenic capacity in the HUVEC assay (Figure-5G).
