Given the central role of NLRP3 activation in IHD, several drugs have been investigated as anti-inflammatory agents in the cardiovascular field; some have failed and been discontinued (atreleuton, canakinumab, darapladib, losmapimod, methotrexate, PF-04191834, setileuton, varespladib, and veliflapon), while others are still in development (e.g., allopurinol, anakinra, colchicine, everolimus, IFM-2427, inzomelid, montelukast, somalix, tocilizumab, and ziltivekimab). This evidence concerns the gene NLRP3 and myocardial ischemia.
