While viral gasdermins do not contain a caspase cleavage site, inflammatory caspases bind gasdermin regulatory domains at an exosite.28,29 A superposition of eptesipox gasdermin with the gasdermin D CTD from a co-crystal structure of human caspase-1 and GSDMD does not reveal any steric clashes, indicative of a potential caspase-directed role during infection (Figure S2G).
