This conjecture was further confirmed by the subsequent mRFP-GFP-LC3 assay (Figure S3C-D), which showed that following addition of Rapamycin, the number of autolysosomes (red dots in overlays) increased in siNC-transfected cells, while in siSDC1- or siTGM2-transfected GBM cells, only the number of autophagosomes (yellow dots in overlays) increased and the number of autolysosomes (red dots in overlays) remained at a low level, indicating that knockdown of SDC1 and TGM2 inhibited the formation of autolysosomes. This evidence concerns the gene TGM2 and glioblastoma.
