NLRP3 and Alzheimer disease: MCC950 can inhibit the activation of NLRP3 by preventing the hydrolysis of ATP to ADP through non-covalent binding to Walker B in the NLRP3 NACHT domain. MCC950 has been used in a wide range of diseases involving NLRP3, including cryopyrin associated periodic syndrome (CAPS), nonalcoholic steatohepatitis (NASH), rheumatism, Alzheimer’s disease and so on. However, it has been discontinued in clinical phase II due to hepatotoxicity.
