The CROWN trial was a large-scale phase III trial of patients with advanced, ALK-positive non-small-cell lung cancer comparing the efficacy of lorlatinib to crizotinib, which demonstrated a significantly-greater progression-free survival for patients treated with lorlatinib compared to those treated with crizotinib, with no significant increase in the rate of adverse events [84]. Here, ALK is linked to non-small cell lung carcinoma.
