Small molecule EGFR tyrosine kinase inhibitors (TKIs) directed against this receptor, such as gefitinib, erlotinib, and afatinib, have changed the paradigm of care for NSCLC, particularly in patients harboring activating EGFR mutations.8 Although treatment with TKIs shows superior efficacy compared with traditional chemotherapies in managing LM,9-11 development of resistance to these small molecules is a frequent feature of disease progression. The gene discussed is EGFR; the disease is non-small cell lung carcinoma.
