The connection between FOXP3 and autoimmunity is well established: mutations in FOXP3 cause a systemic and severe autoimmune syndrome in humans called immunodysregulation polyendocrinopathy enteropathy X-linked (IPEX) syndrome (Bennett et al., 2001; Chatila et al., 2000; Wildin et al., 2001) and the equivalent scurfy in mice (Brunkow et al., 2001).
