While the inhibition of IRAK1 does raise concerns regarding immunosuppression and secondary infections, clinical trials of pacritinib for autoimmune diseases, fibrotic diseases, and oncological diseases have shown acceptable safety profiles [87], and pacritinib has been FDA approved in the USA for the treatment of myelofibrosis marked by severe thrombocytopaenia.
