This uncommon β-sheet conformation for LIR motifshas only been reported for a few cases, including the FNIP tumor suppressor,33 the Legionella effector proteinRavZ.32 A cursory search of reported LIRstructures suggests that ALFY likely contains a β-sheet embeddedLIR motif.49 We provide conclusive evidencethat the bent β-sheet conformation for LIRATG3 isrequired for efficient binding to LC3/GABARAP and is not an artifactof cocrystallizing LIR peptide and LC3/GABARAP. The gene discussed is MAP1LC3A; the disease is neoplasm.
