Interestingly, the dCas9‐KRAB nucleated heterochromatin formation extended locally, with ≈9000 bp downstream of the ZEB1 locus and ≈12 000 bp upstream of the ZEB1 locus, into the promoter and coding region of the neighboring gene, ZEB1 antisense 1 (ZEB1‐AS1), which encodes a long‐non coding RNA that positively regulates the expression of ZEB1 in hepatocellular carcinoma[65] and prostate cancer models.[64] Thus, it is possible that the formation of repressive heterochromatin at this locus could also re‐enforce ZEB1 silencing catalyzed by dCas9‐KRAB in the edited TNBC cells. This evidence concerns the gene ZEB1 and Familial prostate cancer.
