To further confirm the effects of the pathway on IL-17A–induced colorectal cancer cell pyroptosis, we selected the two targets with the highest inhibition efficiency, si-IL17RA (Additional file 1: Fig.S2) and a caspase inhibitor (Z-YVAD, a specific inhibitor of caspase-1; Z-LEVD, a specific inhibitor of caspase-4; and VX-765, low-dose caspase-4 inhibition at a low dose, caspase-4 and caspase-1 inhibition at a high dose), as well as the mitochondria superoxide inhibitor Mito-TEMPO to analyse the pathway of IL-17A-induced pyroptosis (Fig. 6E).
