MAOB and Parkinson disease: Safinamide is an orally administered alfa-aminoamide derivative, currently used in the treatment of fluctuating PD, given its dual mechanism of action: it selectively and reversibly inhibits monoamine oxidase-B (MAOB), and modulates glutamate release: it reduces subthalamic/nigral glutamatergic hyperactivity through use-dependent sodium channel blockade, which prevents calcium channel opening (Pagonabarraga et al. 2021; Stocchi et al. 2022).
